The use of DNA codon randomization techniques enables the substitution of defined positions in the core peptide with all 20 standard amino acids. Employing the randomization exclusively to non bridge-forming residues will keep the original structural framework intact. After codon randomization of the precursor gene each variant will be translated into a variant precursor peptide with unique sequence. As the leader peptide is unchanged the modifying PTM enzymes will recognize the novel precursor peptide and perform all biochemical modifications on the core peptide resulting in a new variant of the deployed scaffold.
